Ya tried but still it gets precipitated when i add in tyrode solution. Can you tell me which organic solvent is to be used and how to avoid precipitation in tyrode solution.
The trick is to first make a mildly alkaline FA solution in alcohol so all carboxylic acid groups are protonated and then dry it. Redissolve it in Tyrode, at high temp and add your BSA while hot.
I've attached a protocol and the ref. we used it in.
Yeah, evaporation of alcohol must be complete before it comes into contact with BSA, as that will denature it. We only did the first step because we did very high solutions, perhaps you can skip it.
I add equal volumes of water and ethanol to the sodium palmitate and boil the solution with stirring until the alcohol evaporates. Mark the water level on the beaker or conical flask before adding ethanol so that you know when to stop boiling of the solution. Although the solution is clear when hot but becomes cloudy on solidification. Hope this helps you.
Palmitic acid will dissolve in ethanol but it takes too much time.. instead you can add Tertiary Butyl Methyl Ether 50% and then make solution with adding 50% ethanol...
Palmitic acid is soluble in all of these solvents n-hexane, Ethyl acetate, Dichloro methane, Acetonitrile, Methanol, ethanol according to increasing order of polarity.
