Hi everyone, after staining the leaves with NBT dye, i have to analyse the result with Fiji. I was stuck at leaf blade extraction. Can anyone provide a step-by-step procedure? Any other imaging software is also acceptable. Attached below is one of my result samples. Thank you.
Your question is miss leading, what you're actually asking is about image analysis, your treatment is ROS related.

Instead, the question should be "Does anyone know how to analyse the picture of NBT-stained leaves?"
Anyway, thank you for your suggestion.
I suggest first you got to bleach the NBT stained leaves to elimnate chlorophyll and then go for imaging as suggested by Richard.
Bleaching process:
After staining, the leaves were then cleaned in a 9:1 mixture of ethanol and glycerin at 70 °C to eliminate the background green color completely. Samples were stored in 70% glycerol for microscopic observation.
