Can Seeds directly transformed by agrobacterium to give rise to transformed seedlings after germination? Whether the plants will be chimeric or transformed or the Seeds of progeny plants having the gene of interest but not the T0 plants?
To my knowledge flower dipping doesn't work with legumes in contrast to Arabidopsis thaliana. We use the protocol published in the Lotus handbook for transformation of Lotus japonicus using hypocotyls. Particle bombardment of embryos may work. You should check the relevant literature.
Actually I was referring to the following articles:
DOI 10.1007/s00299-013-1467-5(Agrobacterium tumefaciens-mediated in planta seed transformation strategy in sugarcane)
DOI 10.1007/s12010-013-0359-z (Assessment of Factors Influencing the Agrobacterium-mediated in planta Seed Transformation of Brinjal ( Solanum melongena L.)
These people have taken seed directly for treatment and produced transformants.
I am trying the same with chickpea and pigeon pea.
