I know that direct smears works well for detection of pathogens in hemolymph, as the technique described above. This work have done midgut direct smears (http://link.springer.com/article/10.1007%2Fs00436-009-1697-1), but without success on detecting pathogens. I have never seen direct smears with salivar glands, but I think you can try by squashing slides with glands collected as the technique described above, with subsequent staining.
