Microsporum gallinae grows well on most nutrient agar. I use potato dextrose agar supplemented with yeast extract and culture at 28-30°C. The Microsporum colony when visualised from the top is a white, satiny colony. It initially produces a yellow pigment which is visible from the bottom of the plate and at the growth edges. As the colony grows and produces micro and macroconidia the pigmentation intensifies and turns red. Conidia production can be enhanced by the addition of thiamine or yeast extract to your media. Keep in mind that the red pigment is most visible when viewing the culture from the bottom of the plate. Mature colonies with good pigmentation are best achieved by a single small inoculum placed in the center of the culture dish and cultured for 1 - 2 weeks.
