I am using RO-3306 CDK1 inhibitor drug to get IC50 and my cell lines is stops responding to to the drug and reach IC50 as previously, any suggestions? or other way to count cell death (I am using now Alamar-blue)

The best way to count cells in drug assay is by using Annexin V & PI staining with Flow cytometry. MTT, WST, CCK8, XTT, Alamar blue assays tell us about cell viability, not actual cell proliferation. One simple way to do a trypan blue assay where you can count the Live/dead cells based on dye exclusion. BrdU or EdU is more evident to understand cell replication.

Saurabh Mandal I used Alamar blue previously and counting live/dead cells as Saurabh suggested, I will read about Promega and check other methods. Thank you so much your answers have given me good insights.
