Hello dear colleagues! Im looking for a simple method to remove platelets from blood samples. Gradient centrifugation doesnt work well because platelets have same density as mononuclear cells, and i need all fractions of leukocytes. Maybe there is solution that selectively lyse platelets or some other cheap methods?
Dear colleague, Platelets does not have the same density as mononuclear cells a simple method is spin the blood at 200xg for 10minutes to separate RBC and mononuclear cells from the PLATELE T RICH layer and aspirate supernatant
Sorry, not the same but similar density so they enrich mononuclear fraction during any centrifugation method (except using ficoll 1.060). There are a lot of publications showing that simple centrifugation and washing steps dont allow to completely remove platelets and their residual number varies too much. But its hard to reproduce their methods too.
I think that the method suggested by Emmanuel could be not suitable for the requested purpose. In figure 1 of this article, many leukocytes are present together with platelets. It seems to me that the missing of leukocytes should be avoided or reduced
