Dear Colleagues,
During in vitro maturation of "camel" cumulus-oocyte complexes, I frequently noticed that cumulus cells start to attach to IVF non-treated dishes within few hours of culture.
I have not observed this attachment during my IVM work in bovine and porcine oocytes.
I think this attachment affects the quality of oocytes.
Do you have any explanation for that?

Some of the OCCs from unhealthy or atretic follicles tend to show such cell attachment instead secreting hyaluronic acid to allow the cumulus expansion in all species including human. The cells may also derived from early non-competent preantral follicles due to the pressure of rare sample collection by sacrificing right follicle selection.
I had the same problem with goat oocytes. You could try adding serum or BSA to the IVM media to prevent the attachment. Also check the type of culture dishes that you are using!

Thanks Maajid for your answer. My IVM medium contains 10% of either FBS or follicular fluid and the same happen when using any of them. Also, culture dish is used for routine IVF (no-coated).
Dear Islam,
I would recommend you to check the CO2 concentration in your incubator and the pH of your media. In general, somatic cells (like cumulus cells and fibroblasts) like to grow in slightly alkaline media (pH around 7.5 - 7.6). Maybe by just adjusting your culture conditions you can prevent them from growing so much and also your oocytes will like it better.
Thanks Dr Carolina for your feedback. You are right, two days ago, I just found a problem in the gas sensor of the CO2 incubator and currently we are fixing this problem. I will update my results after fixing this issue.
