In this case ribbon sections can be floated on the diluted alcohol (1:10 to 1:15 with D/Water) solution by putting on slides itself or coplin jar for few seconds to 2 minutes and then transferred to the water bath. From the water bath, the sections can be taken on an adhesive applied glass slides.

I think there are two possibilities for getting curve sections.

1. you should check the alignment and angle of your microtome knife with tissue. and 2. Make sure that your tissue block has uniform surface, sometime the difference in the thickness between left and right side led to curved ribbon with wrinkled tissue.

If the alcohol method doesn't work, and you have a block that you no longer need, try trimming the block face before sectioning so that it is smaller than what you show here. That way, there will be less paraffin around the tissue. When you float the ribbon, the tissue should have more room to expand. Sometimes one has to carefully make slits in the paraffin using a needle probe so that the tissue can expand even more. This is difficult to do well, so again practice on tissue that you no longer need. You may need to steady the section with a second probe while you do this. I do not have my old technique books that show this, but you may be able to find them in your science library.

Try poly l lysien coated slides which adher the sections well! Check if you can increase the tempreture of water bath

Hi Ruth Kelly , did you find a solution to the wrinkles problem? I tried the ethanol bath trick and seems a bit better. I work with hydrogels, a water-rich material that is very hard to process for paraffin embedding!
