My group works on isolated smoothmuscle strips and in isolated rat kidney. Anybody who has experience in the use of isolated and perfused rat kidney?

Concerning perfusion, you can perfuse the hole blood system.

To perfuse brain, we are using first PBS to wash the system and then 4% PFA. People are using around 200 ml of PBS and 200 ml PFA.
