I am trying to make amino acid standard curves for HPLC analysis and I am going to use a pre-column derivatization with OPA. My question is how stable are the OPA-derivatized amino acids? Can I derivatize first then make serial dilutions from the derivatized amino acids without degrading the derivatized compounds? Your inputs would be appreciated.
I suggest you to proceed analysis by postcolumn derivitisation with flourascent detection. Because OPA derivitised alkyl thiol iso indol derivatives are not stable for a long time and elution time also to be considered. Pls find reference articles in the attachment
Hello go through my paper, opa derivatives are more than 3 hours they're stable it is go enough to complete the gradient separation within 45 min.
