Hello,
I have read in one paper that to ensure proper dissolution of the formazan crystals with DMSO, the plate must be air dried overnight before DMSO addition.
Is it true that it may be difficult to dissolve the crystals and that air drying could help? I am afraid the crystals are not stable after one day even if the plate is air dried in the dark.

It is not difficult to dissolve the crystals. Do not air-dry the plate. You will not get proper results. Just follow the protocol below.
After adding MTT to the wells, incubate the 96-well plate for 1-4 hours whichever time point you have optimized. Aspirate the medium with MTT from the wells. Add 100ul of solubilizing solution (DMSO or SDS or acidified propanol) to the wells and incubate for 15-20 minutes at 37 degree C to dissolve the formazan crystals. The formazan crystals are easily dissolved.

Hi Sandra López Cerdá I do agree with Malcolm Nobre It can be done by just removing the medium with MTT from each well and then adding the solubilizing solution. I did the experiments few times with consistent results.
