I am currently in the process of setting an acceptable criteria defined based on total allowable error (% deviation), for inter-laboratory comparison of the following assays performed on human cord blood samples:
Total Nucleated Cells (Sysmex XN-series)
Nucleated Red Blood Cells (Sysmex XN-series)
White Blood Cell Count (Sysmex XN-series)
Mononuclear Cells (%)
% CD34+ (BD FACSCanto/II, ISHAGE protocol)
Total Nucleated Cell Viability (%)
CD34+ Positive Events
Absolute CD34+ Count (/µl)
% of CD34+ within CD45+ Population
CD45+ Viability
CD34+ Viability
I would be grateful if you all could advise whether CLSI, ICH, ISHAGE etc provides specific guidance or acceptable limits of variability for these parameters, for such assessments. Any references to share?
