Giemsa stains the proteins of the chromatin. The R banding is achieved by differential changes in the chromatin structure due to the chemical temperature treatment of the chromosome material. Several text books and papers describe these processes in detail ( e.g., Sumner AT. Chromosome banding. 1990)
