Hi dear all,

I have started to do lipid extraction from microalgae. It is my first time to do this experiment. I used Folch method and hexane:isopraponal method..

I collected microalgae and dried at 60C. I powdered and 20 mg algae powder used for lipid extraction..I added 6 ml chloroform:methanol mixture and forward steps made according to Folch method. Lower organic phase was clear yellow. My question is that is it normal to use 6 ml solvent mixture for 20 mg dry sample?

Because literature suggest 1 g sample in 20 ml of solvent mixture..On the other hand, I used 1 ml solvent mix. it's very dark lower phase..(fig 1.)

Also i used 1ml hexane:isopropanol mixed with 20 mg dried sample is very clear yellow lower phase.
