Can liposomes be formed using DPPC lipid only in acetate buffer pH 5.3?

I made liposomes using Ethanol injection method, I try DPPC (2.5, 3.4, 5 mM) in acetate buffer. I fixed the volume ratio of Etoh : buffer (pH 5.3) to 1:20 but i think No Liposome were formed...

How can I calculate the %yield of chitosan nanoparticles especially if I don't have a freez dryer or ultracentrifuge?

I made chitosan nanoparticles. I centrifuge and nanoparticlesill be on the supernatent. The concentration used 0.75mg/ml chitosan and 0.75 mg/ml TPP all formula 15 ml in acetate buffer 0.1 M.
