The DNA could be either PCR generated or a product of RE digestion. Also, if there is any particular minimum limit to the length of the target DNA to be self-ligated? Thanks.

In particular, my target is Taxus spp. So far, i have tried Sigma kit as well as TriReagent Methods but unsuccessful. Have seen some literature also but the protocols there seemed a bit complicated.
