At which different wavelength various components of bone amide 1 will appear while taking second derivative of amide 1 ?

I need to know at which wavelength random coils , B-sheets , alpha- helix etc will appear in amide 1 of bone using raman . Lot of literature is there for FTIR but i couldn't find for RAMAN.

While doing raman spectroscopy of bone sample , which peak or peaks should be used so as to quantify amount of collagen matrix?

There are many peaks of amide in the spectra, which should i consider?

Is there a way to find localized Poisson's ratio of bone using nano-indentation?

I am trying to find a way of using load - displacement data to find the localized poison's ratio of bone. Literature says we generally take 0.3 for cortical lamellar region but can we validate it.
