Hi, I want to clone the full length of potyvirus which is a positive single stranded RNA with a size of 9.7 kb. How can I clone it?

Since the size is a bit larger (9.7 kb) I have to clone it in fragments (subcloning). Overlap primers can be designed for PCR amplification.

Why do we potyvirus clone in fragments (by subcloning) and not full length genome at a time?

I think the genome size of potyvirus is ~10 kb but how does this large size hinder cloning? How can we construct an infectious clone of potyvirus?
