Hi , I would ask you if somebody have used less than 200U in the RT-PCR reaction?

I want to save more enzyme that´s why I want to use 100 or until 50 U in RT-PCR reaction. It that possible without any change in the amplification? I have used enzyme of Invitrogen and Ambion

Does anybody know if CMV promotor in a DNA plasmid can be methilated? because there is not more expression of the target protein in mammalian cells?

We had tested the expression of EGFP proteín in mammalian cells, before we got fluorescence, but currently the fluorescence resulted inexistent. thanks a lot for you comments
