I would like to take bacterial OD600 measurements on a microplate reader? How can I do that?

I understand the OD measurement in a plate reader varies with the volume of the sample measured. Is there a formula to correct for path length without finding the correction factor by myself?

Is pET21 vector compatible with Rosetta host cells?

pET21 contains ampicillin gene as the selectable marker and Rosetta cells require Cloramphenicol because of the pRARE plasmid? What antibiotic should I use then?
