Has anybody tried applying plant extracts on MCF 7 cell lines after filtering them? will it (filtering) effect the antioxidant activity present in plants?

For DPPH assay, Why we always 0.004% soln of DPPH in methanol and set 0.7 to 0.9 as an ideal absorbance value to work with?

how can we justify that we need 0.7 to 0.9 ideal absorbance values before putting it into test samples.
