How many times should the permeability of the transwell-membrane covered by endothelial cells differ from the uncovered membrane?

To model the BBB using transwell-membrane, I use bEnd.3 cells. The permeability of the monolayer is assessed by bypass of the fluorescent tracer Na-fluorescein through the monolayer.

Why do bEnd.3 cells stop forming monolayer and change morphology after long freezing?

Mouse brain endothelial cells bEnd.3 have changed their morphology and do not perform ability to form a fully integral monolayer. It happened after 1,5 years exposure to freezing in nitrogen vapors.
