I would like to use IR spectroscopy on dentin slices of 1 mm thickness . i would like to investigate the samples from one surface to the other. Thus, I need to know what is the maximal penetration depth (Z axis) allowed by this technique to analyse the spectrums.
At 1mm thickness you are unlikley to be able to do transmission IR spectroscopy. If you move to ATR with a diamond window you are typically looking at pentration depths of 2 - 5 microns (which is wavelength dependant). This assumes good contact with the diamond. You may want to use an array detector to create chemical images from the area measured rather than a bulk spectrum with a standard ATR accessory. Whatever your approach, there will be challenging practical aspects of this analysis.
Is there any specific reason for using IR? Raman may offer some advantages for Z axis analysis.
Look at the work from Michael Morris' group (Uni Michigan). That might give you a starting point for the type of analysis you want to perform.
