following composition of YEB works very fine

D-Mannitol: 10%

CaCO3: 4 %

K2HPO4: 0.5%

yeast extract: 0,4%

MgSO4.7H2O : 0.2%

NaCl : 0.1 %

pH 6.8-7.0

for Agrobacterium you can used Yeast Manitol Broth/agar, or Luria Bertanii for miniprep plasmid

i suggest you using YMB,

YMB compositions are

Yeast extract : 1g/l

K2HPO4 : 0.5 g/l

MgSO4.7H2O : 0.2 g/l

NaCl : 0.1 g/l

D-Mannitol : 10 g/l

distilled water 1 liter

but if you want YEM (do you mean YEM is yeast extract mannitol medium ?, if yes i give you the recipe taken from handbook of microbiological media)

Media - YEM (YEast extract - Manitol)

YEM: Composition: Mannitol: 10g, yeast extract: 0,5g, K2HPO4: 0.5g, NaCl: 0.2g, MgSO4.7H2O : 0.1g CaCO3: 4 %

distilled water

pH 7.0

Prepare 16.6% CaCl2 solution.

Autoclave for 20 min at 120 °C

Add 1 ml of the 16.6% CaCl2 solution to 1 liter YEM medium (1:1000)

Let cool to ca. 55 °C before adding antibiotics

Store at room temperature or 4 °C

we are using 10g BactoTMyeast extract, 10 g BactoTM Peptone , 5g sodium chloride for 1 litter volume and adjusted to 7.5 pH. Finally we autoclave and use after cooling or store it at 4°C until use.
