In betalain biosynthesis pathway, formation of L-DOPA from tyrosine is the first step. I silenced a gene, which converts L-DOPA to further products and I also observed the expected phenotype on the leaf pigmentation, but I didn’t get the accumulation of L-DOPA on leaf tissue, as expected. Could you please share your thoughts?

In a biosynthetic pathway multiple enzymes and multiple substrates( each product of the enzymatic pathway substrate for for the another enzyme).
Hence, till reach the ultimate end product there is a rate limiting and feedback inhibition( each step). if you stop conversion to next step it will inhibit its own production by inhibition of previous step enzymes at various rate.
Inter cellular reaction is continuous and multiple, so that ultimate end product also have its own level of concentration in a cell. moreover, ultimate end product may weakly bound with other molecules like proteins so it may not available for its own inhibition until some point of concentration.
1. color change achieved due to non formation of betalain.
2. l dopa non accumulation may due to feedback inhibition.
this might be identified by in vitro studies with various concentration of L dopa.
