Did anyone observe that semisolid MS medium becomes liquid after a few days for culture (Incubation with plans at multiplication stage)? I have plant culture (Coleus forskohlii) in the multiplication stage; I have subcultured it onto MS Basal semisolid medium; after a few days of incubation, the semisolid medium turned into a liquid medium.

I would like to know what are the reasons behind these changes..!

I wondered if my plant is producing secreting some chemicals which convert semisolid (Agar-agar degrading) medium to liquid medium...!..?

pH change may be one of the reasons for this; what is making this change?
