In kang kong, plants are raised both from seeds or cuttings although plants raised from seed is the normal practice. But it has been observed that not all the stem cuttings of kang kong used for propagation exhibited desired performance i.e. produced healthier plants. Only one or two specific nodes exhibited better performance.
Dear Sitesh Chandra Biswas there're several reports on the selection of explants sources for determined purposes. Indeed you have provided scarse details about what's happening with your unsuitable explants. What does mean "better perfomance"?
From cuttings I wanted to develop complete plant in kang kong but only the 7th and 8th node of a vigorously growing branch from the base and 4th and 5th node from the shoot tip produced complete healthy plants and these healthy plants turned in to full grown plants which exhibited excellent vigour, produced huge no. of flowers and fruits which I described as 'better performance'. But except these 2 nodes, when other nodes were used as cuttings did not produce good plants.
As I told you, the selection of suitable explants not only affect to your plant , but it's also frequent for other species. In fact, the election of right explants is considered among the main factors with great influence on the final result of in vitro culture. Lignification degree, ontogenic age, accumulation of some metabolites (carbohydrates, phenolics...), among other, are considered factors that might determine which explant is suitable, or not, for that are you looking for.
In general, the explants collected from apical meristem are disease free. Hence specific nodes may be selected for successful clones.
Well, this is true for tissue culture technique but which node or nodes are suitable for raising healthy plants when 'Cutting' technique is followed in case of vegetatively propagated plants?
