As usual i am checking my Agrobacterium strains with cabbage, carrot and potato. But in potato i am seeing it turns black color. Is it because of oxidation or some thing else? Kindly help me in this regard.
Which part(s) of potato you are infecting by Agrobacterium and trying to transform ? Potato infected explants may respond differently by secreting some phenolic compounds as a part of defense mechanisms

1. I think that it probably caused by phenolic compounds. I saw this same phenomenon on cotton transformation. It became dark around the cut explants. But, I don't have experience on potato.
2. By the way, what material did you use for transformation?
3. I attach a 'potato transformation slide' for you. They use leaves. You can estimate the size/age of the leaves (with the petri-dish) they used for genetic transformation. You might be able to get an idea of how other researchers perform potato transformation.
Good Evening! Hope all your health is good. May be Phenolic compounds are oxidizing in my case. I am using direct potato ( Cut into small pieces) and giving infection for 20 min with Agrobacterium strains.(Half Strength MS Media)
But Interesting one is i am doing my experiment to just check weather the strains are in good condition along with my seed ( that is i am giving infection to seed also along with potato) But i am not finding color change in my seed. Is seed not release phenolics into media?

Its better to use in-vitro maintained shoots for explants. we always use internodal cuttings of 5mm and routinely transform using Agro. Potato tubers have lot of bacteria in them and could lead to contamination problem.
