Hello, Sunil! Bacteriological method regard as golden standart in microbiology. Micobacterium tuberculosis is fastidious bacteria and do not grow on common media. That is why LJ media which has lots of additional nutrients is used for cultivation of Koch's bacilli.
Culture of Mycobacterium tuberculosis complex from patient specimens on solid growth media, such as the LJ slope, remains a gold standard in the detection of active tuberculosis due in part to its relatively high level of sensitivity with respect to other available TB diagnostic tests. It is low cost enabling it to be used in low-resource settings and it provides colony material for subsequent phenotypic drug susceptibility testing. The downside is that MTB can take 3 weeks or more to produce visible colonies on LJ slopes. Normally, other diagnostic techniques are performed in parallel during this time. But the inclusion of the LJ slope, or another solid growth medium, in TB diagnostics is still recommended by the ECDC and other organisations.
