Hello Everyone!

Yesterday i was analyzing 13 Plex by using Luminex Magpix Instrument. In the morning the Cal & Ver was failed, I sonicate the probe, perform enhance statrup routine than Cal & Ver passed.

After a while i tried to acquire my 13 plex assay but the not bead count were showing in any of the wells although the sampling was done by following the protocol. again i tried to acquire that plate only two wells have the good bead count rest all were no bead count. later i perform Di Water test no beads were showing in the result means there is no clogging in sample path and after Di Water test i run Ver as assay agaiun the bead count was too good.

by today i run again verificatiop performance after probe sonication and daily fludics prep and it was successful. Than i tried to reacquire my same 13 Plex plate, now the bead count was very good even in those well where no beads were showing yesterday.

My questions are:

What could be the possible reason for no bead count?

Does drive fluid takes any role in bead count?

Does is there any time boundation for expiration of drive fluid if it is in instrument?

What step i can take for removing the clog in the sample probe othe than sonication?
