Many reports show 60-100 mM KCl-induced vascular contraction of maximum. In my experiment, I try to induce thoracic aorta contraction by 60, 80, and 100 mM KCl for 3 times. The results show that the second or third time have maximum response. Then I conformed with 1x10-4 M NE for maximum contraction (prior in lab, the cumulative concentration response to NE show maximum response at 1x10-4 M). But this response is unequal to induce by 60-100 mM KCl. The maximum contraction by NE show response more than KCl-induced. Actually I want to use KCl to induce vascular contraction to normalize the tension response to NE. When normalize its should less than 100 % maximum of KCl.
Please discuss in my experiment.
Actually we are working on human tissues as saphenous vein and also induce KCl 40 mm contraction and later on noradrenaline dose response 10-8 - 10-4 , it happened that usually it was also higher 100% and also lower when normalised contraction from contractions from noradrenaline. So it depends but also you need to take in consideration that normalisation with 1 gram for thoratic aorta. Hope to find this information good from you.

The optimum of resting tension for thoracic aorta rat in my experiment is 1 gram. I tried many concentration (30, 60, 80 and 100 mM KCl) for induce contraction. All concentration of KCl induced contractile response less than 1x10-4 NE. Actually I want to avoid normalization from resting tension because sometime the tissue did not good representability response. I know the force depend on length and surface area of ring segment. From this results, I will adapt to yield higher 100% than maximum response to KCl.
Yes you can do like that. As we are doing same in other tissues. So important is to have a control as you better know. We have results either under 100 % or up to, so take care also to have good preparations and equilibration time until is stable approximately 1 g.
In line with the main question; I have been carrying out some studies on the Corpus cavernosum of rats and I have been experiencing some challenges. Please Can KCl totally substitute Phenylephrine OR NE in precontracting or contracting the tissue. If Yes... Is there any recommended range for the dose... I have not been getting any meaningful results with the doses I have looked up in journals.
