I am trying to observe the oil body density of aleurone cells in mature rice seeds. I tried embedding it in Technovit 7100 resin using the protocol published by Ryo Matsushima (http://www.bio-protocol.org/e1239). I used a glass knife to section the resin blocks at 2µm thickness. However, the specimen turned into a powder-like consistency when sectioned.
Are there any possible solutions regarding this problem, maybe a modification in the protocol or cutting method?
