We are attempting multi-patch clamp recordings in cerebral cortex but are experiencing severe blockage issues. Our pipettes are frequently clogging.

Our internal contains: K-gluconate (120 mM), KCl (10 mM), HEPES (10 mM), Na2ATP (4 mM), NaGTP (0.3 mM), Na2-phosphocreatine (10 mM)

All ingredients are new and bought from Sigma. The internal is pHed to 7.3 with KOH, the osmolarity is 295. It is filtered with a 0.2um filter after being made before being stored in a -80 freezer. Before use it is thawed and filtered again with a 0.2um filter.

Is it possible that something could come out of solution / crystallize over time to cause the blockages? We're relatively certain the problem isn't our AgCl electrodes, pipette holders or pipette fillers.

Has anyone experienced this sort of issue before with K-gluc internals? Any advice would be very much appreciated.

Many thanks,

Joe
