We started the culture of human coronary artery endothelial cells (HCAEC) for a few days. The cells multiply rapidly (2 passages per week), but have many intracellular vesicles visible in light microscopy. Have you ever had a similar situation with the same cell type or a close type? Thanks for your help.
Hi, what is your concentration of serum in medium? As they are multiplying rapidly I would think, that the levels are high.
The high amouth of intracellular vesicles may be caused by high levels of serum (as they are dividing rapidly, they are eating rapidly :) . This is one of possible explanation, but it is hard to say without picture, cumulative population doubling of cells, serum concentration and question is if it is really too many...
I have seen intracellular vesicles in HAoECs (aortic), HUVECs (umbilical vein) and THP-1 and they were in most time caused by high levels of serum (higher amouth of vesicles directly after thawing of cells lower in later passages).
