I'm working with a plant and have gone through methanol and hexane extractions (both crude extracts have been obtained). I have a hexane insoluble layer, which is supposed to be extracted with a (1:1) ratio of ethyl acetate/dH2O. But instead of getting 2 separate layers, I have one homogenous, clear solution. Where did the ethyl acetate and dH2O layers gone to?
It is happening due to similar polarity, please use solvents of two opposite polarity or non polar to polar, you will get sharpness and clarity in separation. Please change the ratio and try 1:4 ratio.
Please do not use to miscible solvents, always use contrasting polarity bearing solvents for your use. It will separate in butanol or in ether. Please try.
For sure do not use mixture of solvents in extractions. Try with diethyl ether and water and shake gentle. Avoid the formation of emulsions and put a with paper behind the separator funnel. Somtimes separation of layers is barely visible.
