Why can't I see any bacteria under my experimental inverted microscope setup?

Hello,

I've set an inverted microscope setup on optic table to see bacteria on 0.13-0.17 cover slip with short WD objective(0.17) but I just could see them once and have difficulties to regulate my focus and it seems unrepeatable.

Could you please tell me what I should do to have adequate image of my sample?

Dear Mohammad,

Could you please tell us a bit more about your set-up? What is the staining for your bacteria? Medium? Do you want to use transmitted light or fluorescence?

If you cannot regulate you focus, maybe your bacteria are "too far" from the coverslip (regarding to the WD). Maybe there are some things to try in the way ou mount them on the slide?

I think you know that but do not forget to flip your slide to put the coverslip on the objective side.
