Which one is best method for biofilm extraction?

Which method is best for biofilm extraction. i have used 3 methods for extraction 1- simple centrifugation 2- centrifugation and ultrasonication 3- chemical method using formaldehyde and NaOH. how can i say that which one is best metod?

I want to extract biofilm from metal coupons exposed with inoculated media and want to estimate carbohydrates, proteins lipds etc.
