a. Mycoplasma ribosomal RNA (rRNA) detection by nucleic acid hybridization using a luminescent probe.
b. PCR of mycoplasma DNA.
c. Flow cytometry based on DNA content.
All methods mentioned by you are very sensitive and reliable.
Cheapest and easiest way is PCR.
