When digesting metal nanoparticles synthesized from apoferritin we obtain very different results in ICP-MS measurements. The digestion is made with aqua regia (HNO3:HCl 3:1) and H2O2 (2:3,) for one hour at 60 ºC. Is there any other method that provides a more accurate digestion? Thank you
You can check EPA methods such as 3050B or 3051 for metals digestion.
Microwave digestion at ~180 C is suggested. Before microwave digestion, please lay in fume hood more than one hour and make sure most gas overflow.

I think the digestion part of your method is correct as long as everthing is in solution. I feel the problem is with the analytical method used. I believe ICP-MS is the wrong technique since you are dealing with higher concentrations of the analyte in question (> 500 ppb). You must have diluted extensively to get an answer. The more one dilutes, the greater is the error introduced. I would suggest ICP-OES with proper standards, controls and the addition of internal standard as well as replicate analyses so as to get reasonable results. In most cased no dilution is required.
