I would like to macerate the plants with organic solvent but I have the choice between extract from polar to nonpolar or nonpolar to polar organic solvents. Which one is better?
Extraction of polyphenols from plant extracts is something my research group does quite frequently. We usually macerate exhaustively in 80% methanol first, flash evaporate the concentrate then freeze dry it. You may want to consider removing chlorophylls from your sample by mixing with dichloromethane (before flash evapourating)
But what you need to remember is that the extraction solvent used really depends on the target compounds you wish to isolate.
