In my experience, TRIZOL reagent works better than the kits in terms of yield. However, the purity is better if you use the kit correctly. With TRIZOL, you may have high yield of RNA but contaminated with DNA. if this is the case, I recommend you to do go one step further and use DNAse digestion (Ambion kits works just fine). Good luck.
We've worked with plasmacytoid dendritic cells isolated from preterm cord blood with a minimum cell count of 300 cells. We used the Cells-to-CT Kit from ambion with good results. Another opportunity ist the RNeasy Plus Mini Kit from qiagen. TRIZOL is an effective AND the cheapest variant!

Hey cleetus! Did you mean the RNA yield from pDCs? Between 1- 4 µg RNA with the cells to ct kit, depends on the stimulation. We've investigated the IFNa expression by RT-PCR after TLR stimulation.
@PBMC: It depends on the lymphocyte and monocyte count of the donor. Normally, healthy peripheral whole blood contains 1500-3000 lymphocytes/µl and 280-500 monocytes/µl.
I have only 3ml blood sample.Is it possible to obtain good yield of pbmc and isolation of total Rna from it?@Lukas Wisgrill.
I am in thought to use (Ficoll paque plus/pbs) & (Trizol) reagents for pbmc and total rna isolation.Do i need to increase my blood volume? Suggest me with ur idea.
