To test anxiety and locomotion commonly "Open filed" and "Elevated-plus maze" apparatus are used. However, for certain reasons multiple instruments is not possible to use in same study. To get better idea of anxiety and locomotion which test should be used in the first place and why? What is the benefit of one test over another?

The open field test (OFT) is not quite a good test to evaluate anxiety. OFT is more useful for locomotor measurements than it is to anxiety. Elevated plus-maze is a considerably good test and widely used for anxiety behavior. So you should be very carefull in selecting tests for anxiety. Though reduced ambulation in the centre of OFT may be considered sign of anxiety but the behavior may be confounded by locomotion. You should also consider another behavior test which is better to evaluate anxiety-like behavior such as novelty suppressed feeding or the radial maze to have a better profile of your animals behavior.

Hi Adnan, great! Yea confusion arises here. In some papers it has been naratted that an increased locomotion centre vs periphery and spontaneous activities such as rearing are consodered as anxiety-like behaviour. Thanks for your suggestions and papers. Monokesh

Although the OF test has been used for a wide variety of purposes, researchers most often use it for measuring rodent emotionality (or anxiety or fearfulness). OF thigmotactic or wall-seeking behavior refers to the propensity of a rodent to stay in close contact with the OF wall because of the underlying tendency to avoid open, unknown, and potentially dangerous places (Choleris, Thomas, Kavaliers, & Prato, 2001). The validity of thigmotaxis in measuring emotionality has been well established in mice (Choleris et al.; Simon, Dupuis, & Costentin, 1994) and rats (Treit & Fundytus, 1989; van der Staay, Kerbusch, & Raaijmakers, 1990).
