Which in vitro assays are needed for evaluation antigenicity of a peptide antigen?

Dear all,

I am working on a peptide antigen as a part of peptide vaccine, but due to limitations on laboratory facilities in vivo assays are not accessible. In your opinion which in vitro assays are needed for evaluation of my peptide.

We evaluated the efficacy of epitopes to stimulate T cell responses using ELIspot assays (IFNg) and T2 binding assays. Really you need to know if your antigens leads to killing but that requires mouse immune cells. See Hardwick et al, 2013, for more details.
