We use one of the Ankom Technologies Inc. instruments to analyze the lignin content in compost samples. The instrument uses a type of a filter bag to hold the sample. This type of the filter bag prevents the use of pure lignin powder as a check sample because the lignin powder is too fine, thus coming out from the bag easily.

There are some Reference materials (RM) available where you have a certified lignin content value (NIST , BCR, etc...)
Can this method handle lignin in complex matrices? If so go with NIST 1944 (Hudson River Sediment) or NIST 1547 (Peach Leaves), with published lignin values in Louchouarn et al. (2000). If not, check into the paper manufacturing literature on lignin and cellulose chemical separation methods - you may be able to obtain some standard plant tissue that you can isolate the lignin from and characterize
