I know the best possible explant for transformation in wheat is immature embryos but the problem related to it is, it's too laborious and the time period to isolate it is too limited. Can anyone help me know what can I do if I missed that crucial time for immature embryo isolation. Which explant I can use for transformation except immature or mature embryos in wheat?
As you say immature embryos (actually scutellar tissue) are the best explants for wheat transformation. Another possibility is to use immature spikes (less than 1cm length), but the transformation and regeneration efficiency is lower. In my opinion it's worth to spend the time to gain hands on scutellum isolation rather than using immature spikes.

I did this work many times, it is very intersting and enjoyable but it need more time and Lab possibilities
