The area of my university is more than 150 Acre. It consisting of variety of vegetation patches. Among them dominating vegetation types are deciduous forest patches, plantation area, urban area etc. I want to now what will be best survey techniques to study & stratify vegetation of my university campus.

I would try aerial photography using a drone for such small area
Field work is to be carried out at cadastral level (1: 4.000 scale) considering the slope variation . However the mapping can be done at 1; 10,000 scale using mono-series as the mapping unit.
When the field work is done at cadastral level,the use of aerial photographs may be an essential requirement.
Use the nearest neighbour 100 trees approach where you pick one point randomly and moving in a circular manner pick the nearest 100 trees. Around each tree you can take all the parameters you require
I think you are asking the wrong question intially. You need to be asking what is it you need to know about the vegetation. What are you sampling needs. You design the protocol to sample what you need to know. Is it just what and where is the vegetation? You can do that remotely and then ground verify a sub-sample. If you need to know what it is and how much you have, that is a different question. Start with the need and work to the protocol.
