Yes, it can be hot water soluble polysaccharide (HWSP). The sugar part and molar ratio of various sugar moieties of the polysaccharide can be determined after complete hydrolysis followed by GLC analysis of alditol acetates. The uronic acid content can be studied by assorted approaches. Spectrophotometric methods using chromogens carbazole, m-hydroxydiphenyl and 3,5-dimethylphenol corroborating the results by GLC analysis. Methylation studies, periodate oxidation and other degradative techniques alongwith spectral analysis are used for studying the linkages in polysaccharide chains.

Solubility of HWSPs is indeed a major problem. However, you may dissolving your compound in D2O for NMR spectral analysis.

But it doesn't give complete solubility at normal temperature, then it would be the problem to get real data

try to dissolve it in warm D2O it should not precipitate after cold down, once is cold it should form a viscous solution. Don't make your sample too concentrate. You can register your NMR espectra at 40C to 60C if still you have solubility problems.

thanq @ Roberto Veloso Pita, i tried that way in H2O, but not in D2O, i didnt got any ppt after cooling down. will not get any problem if we warm D2O at atmospheric level, may get H2O peaks in Data?

You are goin to have some H2O peak from your polissacharide, D2O purity etc. You are not going to find much differences due to the D2O warming. You can reduce de H2O by dissolving and freeze drying or liofilizing your sample 2 or 3 times in D2O.

Other thing you can try is to dissolve your polyssacharide in (DMSO)/d6:D2O (6:1) .
