Hi Aaqillah, What do you mean by serial dilution? Why you are diluting your sample. By doing this you are decreasing the total fatty acid content. What do you mean by few fatty acids? is this a diversity fatty acids found in your sample or concentration of fatty acids? And if you are talking about DHA EPA can you specify which technique you used n the analysis. do you mean GC MS or anything like this? And if GCMS then which column you used in GCMS is important.

Tq for your answer. Initially, i did not dilute my sample. But upon submission, they asked me how many times dilution i have made for the sample. I also wonder why do i have to do dilution because all i want to know is the concentration and profile of the sample itself. I'm doing GCMS using a column SLB5MS. When I got the result, it is like what i have expected. There was no traces of EFAs in my sample, i presumed because of the dilution. Can i not doing any dilution? They said, if i do not doing any dilution, the high concentration will result in a damaged GCMS

Yes if your sample contain very high amount of fatty acids then it will damage. But if you have doe literature survey about how much initial amount of sample should take then you can either dilute your sample or reduce initial sample weight or volume. Another thing is in fish its reported to have more EFAs but need to revise your protocol if you are not getting. are you getting any other derivatives of any of them? then you can defend it. but yes, need to see your data carefully for that. and protocol you are using need to be check.
