Moreover, the use you'll give to these semen doses also strongly influence it (for example if you will inseminate synchronized females, vaginal or laparoscopic, transportation to destination, etc.). Diluent Tris and Citric Acid based can even keep at laboratory temperature for a couple of days with acceptable results of pregnancy.
In my opinion, storage of liquid semen under any condition/ extender is better at 5 degree C to attain optimum fertility

If the extender has egg yolk and antibiotics, normally you can storage ram semen for 48 hours without problem. But as I said before, the dependence of the season is important on the results. If you are measuring only lab tests and no fertility test, be carefull with the temperature of maintenance. at 15 C you have several changes on the sperm membrane. You can include the HOS Test that is easy to perform, and can get you and idea of the physiologic condition of it. So, first you have to decide the temperature of maintenance of the samples you try to analyze, and then, determine the protocol to follow. thank you so much for your assessment, So in your opinion this is better that, i storage ram semen in 5 ° C for more than 48 hour? my extender is based on tris and egg yolk and i dont use the antibiotics. as well as i use omega 3,6 and the CLA in my extender.

I have to tell you that it will depend on many things: your breed, the extender you are using, the concentration in the straws (or the spermotozoa number,....), if your extender contain or supplemented with an antioxidant or not, ….. If you can specify what are your conditions I can help more. I have question for you : what is the purpose to maintain semen conservation as long as 4 to 6 days? Because I believe, even you will succeed to do it in vitro the AI rate will be very low.
